I don't have the game, but I am still psyched about the new mission! Anyways, to get it you have to download it via wi-fi. I have the download instructions below:
1. Open EPF and click on "Upload/Download".
2. Click the middle button, "Connect to Nintendo Wi-Fi Connection".
3. Click on the checkmark.
4. After it loads, click on the "Download Mission" button. It should start downloading.
5. Hit the checkmark with your stylus. Now click the left arrow and then the check. Check again.
6. Click "Access preciously downloaded content" on top. Tap "The Puffle Pranksters" and tap the check.
Congrats, guys! You've just begun playing the new mission!
